What is 5-Amino-1MQ?
5-Amino-1MQ is a small-molecule inhibitor of Nicotinamide N-Methyltransferase (NNMT) — an enzyme that consumes SAM (S-adenosyl methionine) and reduces NAD+ precursor availability. By blocking NNMT, 5-Amino-1MQ increases cellular NAD+ levels and supports the methylation cycle, producing synergistic metabolic and anti-aging effects. It has been studied in fat cell models for its ability to reverse adipogenic gene expression.

Key Benefits
NAD+ Elevation
Blocks NNMT-mediated NAD+ consumption, increasing intracellular NAD+ levels and supporting sirtuin activation.
Fat Cell Metabolism
Studied for its ability to reduce fat cell size and reverse adipogenic gene expression in preclinical models.
Methylation Support
Preserves SAM (S-adenosylmethionine) availability, supporting DNA methylation, detoxification, and neurotransmitter synthesis.
Anti-Aging Effects
NAD+ elevation activates sirtuins and PARP enzymes linked to longevity, DNA repair, and cellular health.
Mitochondrial Function
Elevated NAD+ supports efficient mitochondrial energy production and protects against age-related mitochondrial decline.
Metabolic Flexibility
Supports healthy metabolic switching between glucose and fat utilization — a key marker of overall metabolic fitness.
How It Works
NNMT Enzyme Inhibition
5-Amino-1MQ binds to the active site of NNMT, blocking its ability to methylate nicotinamide and deplete NAD+ precursors from the cell.
NAD+ Precursor Preservation
By blocking NNMT, more nicotinamide remains available for recycling through the NAD+ salvage pathway, elevating total cellular NAD+ concentration.
SAM Conservation
NNMT uses SAM as a methyl donor. Inhibiting NNMT preserves SAM for critical methylation reactions involved in epigenetics, detox, and neurotransmitter synthesis.
Sirtuin & PARP Activation
Elevated NAD+ fuels SIRT1, SIRT3, and PARP-1 — enzymes responsible for gene regulation, mitochondrial health, and DNA repair respectively.
